Yes subs can sub in tourneys they run. They usually let the players sub who are in the tourney but have a bye in 1st round of tourney. Thats the way MOST ladders do it, if in 1st round and no other subs are available our TD's will sub.
It's a good idea to keep and eye on lobby in case someone wins that way you can post ur congrats and TY's for playing.

The only thing TD's and HTD's CANNOT do is play and host in tourneys they are running. HTD's may play in tourneys if they are already playing and the TD got booted or had something come up and they couldn't finish tourney out.
